Biography

Janice Aldrich-Wright obtained her BAppSc (Hons) from University of Technology, Sydney and PhD from Macquarie University, Sydney. Subsequently she was awarded the Cornforth Medal, for the best PhD in Chemistry in Australia (1993). From 1993- till present she has been an associate lecturer, Lecture, Senior Lecture, Associate Professors and now a Professor in Chemistry and Deputy Dean of the Graduate School at the University of Western Sydney. She is a fellow of the Royal Australian Chemical Institute and a Fellow of the Royal Society of Chemistry. Taught chemistry but quickly established a strong foundation in research and in this capacity, student mentorship. Her work is internationally recognised in the field of anticancer chemistry and pharmacology and she has published widely. Innovative bioinorganic molecular design, elegant synthesis, comprehensive characterisation (including X-ray analysis at the synchrotron) and the biophysical analysis of the interactions of these compounds with DNA using, for example synchrotron radiation circular Dichroism (SRCD – Aarhus, Denmark), exemplify her research while creating a vibrant, productive and collaborative environment for her research students.

Understanding how molecules find a unique binding position on a segment of DNA ranks among the most fascinating and important current chemical and/or biochemical questions because it is absolutely essential for determining how biological systems work. The rational design of therapeutic drugs design requires an understanding of the physical and chemical principles involved in DNA and RNA recognition and for improved design this information is essential. My research objective has been to significantly extend our understanding of the principles involved in sequence-specific recognition of biological molecules like DNA by carrying out investigations of the binding of metal complexes. The result of this work has far-reaching consequences on the rational design of DNA-based chemotherapeutic agents, effective DNA probes and DNA-sequencing agents. To achieve these objectives a number of metal complexes have been designed, synthesised, resolved and characterised in order to aid our understand of how DNA discriminates between simple molecules by determining their binding affinity and biological activity.

Octahedral metallointercalators are a class of compounds which have been extensively used as probes for DNA recognition. There has been considerable interest, and controversy, in determining their precise mode of DNA binding. Our studies were able to unambiguously demonstrate that these metal complexes could intercalate from the minor groove.

Platinum compounds that are in clinical use, such as cisplatin and its analogues, exhibit their activity by binding covalently to DNA in the cancerous cell thus preventing cell division. The potent compounds we investigate interact in an entirely different way with DNA, through non-covalent interactions. They are highly soluble in water, show very high level of activity in cell lines suggesting they are likely to demonstrate high clinical activity and effectiveness at low dosage and exhibit a high level of activity against cisplatin resistant cell lines.

Microbial biofilm infections play a significant role in chronic infections within the healthcare system due mainly to their resistance to antibiotic treatments. We have determined that our copper complexes exhibit antibacterial activity against the biofilms of Staphylococcus aureus and yeast Candida krusei.
